AKTIVITAS LIPOLITIK PROTEIN LysB DARI BEBERAPA MIKOBAKTERIOFAGA

Abstract


TB (tuberkulosis) telah menjadi penyebab kematian ke sembilan di dunia. TB disebabkan oleh Mycobacterium tuberculosis yang merupakan bakteri kecil yang dapat menular dan dapat tersuspensi di udara. Protein lysB pada mikobakteriofaga dapat menjadi solusi karena dapat memotong ikatan esterifikasi mycolic acid dengan lapisan arabinogalactan pada mikobakteri dan memiliki aktivitas lipolitik. Mikobakteriofaga yang telah diketahui memiliki aktivitas lipolitik, diantaranya D29, Ms6, dan Bxz2. Aktivitas lipolitik pada mikobakteriofaga dapat diukur dengan pengujian menggunakan substrat p-nitropenyl dengan variasi panjang rantai. Diperoleh Bxz2 dengan aktivitas lipolitik paling tinggi (2,7 U/mg) pada substrat pNPB (C4) dan Ms6 dengan efisiensi katalitik paling tinggi terutama pada pNPL (C12) dan pNPM (C14) sebesar 1,14 dan 1,13 min-1µM-1.
